چکیده

Homogeneous preparations of the potassium-requiring yeast aldehyde dehydrogenase are active with either DPN or TPN in the oxidation of a wide variety of aldehydes. Exogenous mercaptan is not a requirement for catalytic activity. The enzyme is denatured in 6 M guanidine hydrochloride to form a protein species with a molecular weight somewhat greater than 21,000. The denatured protein may be reassociated to yield functional enzyme of higher molecular weight.

Purification and properties of the aldehyde oxidases from hog and rabbit livers.

The aldehyde oxidases from rabbit and hog livers have been purified to homogeneity by modifications of and additions to previously published procedures. The final preparations were judged to be homogeneous on the basis of ultracentrifugal, electrophoretic, chromatographic, spectral, and kinetic criteria. A number of physical and kinetic properties of the homogeneous enzymes have been determined...

Rapid purification and properties of potassium-activated aldehyde dehydrogenase from Saccharomyces cerevisiae.

A method for the purification of yeast K+-activated aldehyde dehydrogenase is presented which can be completed in substantially less time than other published procedures. The enzyme has a different N-terminal amino acid from preparations previously reported, and other small differences in amino acid content. These differences may be the result of differential proteolytic digestion rather than a...

Kinetic properties of highly purified preparations of sheep liver cytoplasmic aldehyde dehydrogenase.

The kinetic properties of highly purified preparations of sheep liver cytoplasmic aldehyde dehydrogenase (preparations that had been shown to be free from contamination with the corresponding mitochondrial enzyme) were investigated with both propionaldehyde and butyraldehyde as substrates. At low aldehyde concentrations, double-reciprocal plots with aldehyde as the variable substrate are linear...

Kinetics and reaction mechanism of potassium-activated aldehyde dehydrogenase from Saccharomyces cerevisiae.

Data from steady-state kinetic analysis of yeast K+-activated aldehyde dehydrogenase are consistent with a ternary complex mechanism. Evidence from alternative substrate analysis and product-inhibition studies supports an ordered sequence of substrate binding in which NAD+ is the leading substrate. A preincubation requirement for NAD+ for maximum activity is also consistent with the importance ...

Yeast aldehyde dehydrogenase. IV. Dissociation and reassociation of native and hybrid forms.

Yeast aldehyde dehydrogenase and each of two of its active proteolytic digestion products are readily dissociable into subunits which can, in turn, reassociate. The native protein dissociates into four subunits of equal sire. The partially digested enzymes dissociate into the same subunit plus a second, smaller subunit. The data are interpreted in terms of a tetrameric native enzyme, differing ...
